Actually, once a CF is created and you "apply" it to a queue, all you
need to do is set the CF privileges to let “Whomever” set that value in
the create screen. Navigate thus; Configuration->Custom Fields->(select
CF)->Group Rights. Once in group rights, you decide who can do what for
that CF. Hope this helps.

When did you want this to happen? At create time? Just write a
scrip with the condition to when you want it to happen (may have to
make this user-defined) and then write an action to set the value of
the CF. IF this is the case and you have trouble with the scrip, I
